This biparatopic antibody-drug conjugate (ADC) targets Cadherin 17 (CDH17), a cell adhesion protein frequently overexpressed in gastrointestinal adenocarcinomas, including colorectal, gastric, and pancreatic cancers. Developed by Earendil Labs, the agent utilizes a biparatopic antibody design that binds to two non-overlapping epitopes on the CDH17 extracellular domain. This dual-binding strategy is engineered to enhance binding affinity and trigger more rapid receptor-mediated internalization compared to conventional monospecific ADCs, thereby increasing the efficiency of cytotoxic payload delivery into tumor cells. In preclinical models, the ADC demonstrated selective cytotoxicity across various cancer cell lines and significant anti-tumor activity in CDH17-expressing xenografts.
