birinapant + conatumumab is an investigational combination therapy composed of two agents: birinapant, a small molecule SMAC mimetic that antagonizes inhibitor of apoptosis proteins (IAPs), and conatumumab, a monoclonal antibody agonist of death receptor 5 (DR-5 or TRAIL-R2). Birinapant promotes apoptosis by triggering degradation of cIAP1/2 and inhibiting XIAP, whereas conatumumab stimulates extrinsic apoptosis signaling by activating DR-5. The combination is being studied for relapsed ovarian, primary peritoneal, and fallopian tube cancers under clinical trial settings due to preclinical and early clinical synergy observed with both agents together[1][2][3].
