Bis(N-maleimidomethyl)ether is a synthetic small molecule crosslinking reagent belonging to the maleimide class of organic compounds. It contains two reactive maleimide groups connected by an ether-linked methylene bridge. This compound is primarily used as a homobifunctional sulfhydryl-reactive crosslinker in biochemical and chemical synthesis applications, including peptide and protein modification and the synthesis of DNA/RNA conjugates. In experimental research, it has been shown to covalently modify hemoglobin at specific cysteine and histidine residues, stabilizing hemoglobin structure and preventing sickling in red blood cells—a property investigated for potential antisickling therapy in sickle cell disease[8]. The compound acts as an electron acceptor capable of forming stable covalent bonds with thiol-containing biomolecules[5][8].
