Overview

Bisindolylmaleimide IV is a cell-permeable small molecule that acts as a potent and selective inhibitor of protein kinase C (PKC). Structurally related to staurosporine, it belongs to the bisindolylmaleimide family of compounds used extensively in signal transduction research. Recent studies in pancreatic ductal adenocarcinoma (PDAC) have identified bisindolylmaleimide IV as a novel inducer of calreticulin (CALR) translocation to the cell surface. This translocation is a hallmark of immunogenic cell death, suggesting that the drug could enhance the visibility of tumor cells to the immune system or serve as a mechanism for targeted therapeutic delivery. The compound was identified through high-throughput screening of the NCATS Mechanism Interrogation PlatEs (MIPE) library and has demonstrated the ability to increase surface CALR expression with minimal cytotoxicity in PDAC cell lines such as MIA PaCa-2 and Panc1.
