Bispecific epitope anti-CD5 nanobody CAR-T cells are a novel cellular therapy designed to treat refractory/relapse (r/r) aggressive T-cell lymphomas, including peripheral T-cell lymphomas (PTCL) and T lymphoblastic lymphomas (T-LBL). This therapy utilizes CAR-T cells engineered with a bispecific epitope anti-CD5 nanobody construct, which was developed using a llama-derived nanobody library to target the CD5 antigen. The construct has demonstrated superior cytotoxicity against CD5+ cell lines in vitro compared to previous CD5 CAR-T cell constructs. The therapy aims to provide an effective approach for patients with poor prognosis due to the lack of effective therapeutic options for these aggressive T-cell malignancies. Early clinical evaluation has shown preliminary safety and efficacy, with patients achieving complete remission and CAR-T cell expansion observed.
