Overview

BiTAC-TCE is a conditionally active, dual-targeting T-cell engager (TCE) designed to treat solid tumors, specifically colorectal and lung cancers. It utilizes an "AND-gated" safety mechanism based on the hemibody concept, where T-cell activation occurs only upon the on-cell assembly of two complementary compounds. Each compound binds a distinct tumor-associated antigen (TAA) and carries a split CD3-binding moiety. Productive T-cell engagement and subsequent cytotoxicity (TDCC) are triggered only when both antigens are co-expressed on the same cell, thereby sparing healthy tissues that express only one of the targets. This platform aims to minimize on-target, off-tumor toxicities and systemic cytokine release syndrome (CRS).
