BiTE-UCHT1 is a preclinical bispecific T-cell engager (BiTE) designed to target cell surface heat shock protein 70 (csHSP70) and CD3. Developed by researchers at MD Anderson Cancer Center and Asylia Therapeutics, the molecule utilizes single-chain variable fragment (scFv) sequences from a proprietary csHSP70-targeted antibody and the UCHT1 anti-CD3 antibody. By cross-linking csHSP70-expressing tumor cells with T-cells, BiTE-UCHT1 induces T-cell activation (marked by CD69 and CD71 expression) and subsequent tumor cell apoptosis. It has demonstrated robust anti-tumor activity in preclinical models of peripheral T-cell lymphoma (PTCL) and multiple myeloma, particularly when combined with proteasome inhibitors like bortezomib or HDAC inhibitors like vorinostat, which enhance csHSP70 expression.
