Overview

**BITT-101** is a novel dominant antagonist monoclonal antibody targeting CD40, a tumor necrosis factor superfamily receptor (TNFSR), developed by Boston Immune Technologies and Therapeutics (BITT) using their proprietary DOMab platform. Unlike conventional CD40 antagonists that bind the active trimeric form and can be displaced by CD40L, BITT-101 uniquely binds and stabilizes the inactive dimeric form of CD40, providing enhanced potency for blocking CD40 signaling implicated in autoimmune diseases. It is in preclinical development with NIH funding support for Sjögren’s syndrome, aiming toward an investigational new drug (IND) application, and shows potential for broader applications in inflammation and autoimmunity.[1][4][7][8][9]
