Overview

Bivalent CAR-T (anti-EGFR), specifically the construct known as CART-EGFR-IL13Rα2, is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy. It is engineered to simultaneously target two antigens frequently overexpressed in glioblastoma: the tumor-specific epidermal growth factor receptor (EGFR) epitope 806 and interleukin-13 receptor alpha 2 (IL-13Rα2). By targeting two antigens, the therapy is designed to mitigate the effects of inter- and intra-tumoral heterogeneity and prevent antigen escape, a common cause of resistance in single-target CAR-T therapies. Developed at the University of Pennsylvania, it is administered via intracerebroventricular (ICV) or intrathecal injection to bypass the blood-brain barrier. Clinical data from Phase 1 trials in recurrent glioblastoma have demonstrated feasibility and early signs of bioactivity, alongside manageable neurotoxicity.
