The bivalent filovirus vaccine is a viral vector vaccine candidate designed to provide protection against two high-consequence pathogens: Sudan ebolavirus (SUDV) and Marburg virus (MARV). Developed by the Sabin Vaccine Institute in collaboration with the National Institute of Allergy and Infectious Diseases (NIAID), the vaccine utilizes a monovalent chimpanzee adenovirus type 3 (ChAd3) platform for each of its two components. The mechanism of action involves the delivery of genetic sequences encoding the surface glycoproteins (GP) of SUDV and MARV into host cells. Once expressed, these antigens trigger robust humoral and cellular immune responses, including the production of neutralizing antibodies and the activation of T-cells. This vaccine is primarily indicated for the prevention of viral hemorrhagic fever in regions at risk for outbreaks of Sudan virus disease and Marburg virus disease. It is currently being evaluated in Phase 2 clinical trials in East Africa to assess its safety and immunogenicity in endemic populations.
