Overview

The bivalent norovirus vaccine is an investigational vaccine candidate designed to protect against norovirus infection, targeting the two most prevalent genogroups responsible for human disease, GI.1 and GII.4. This vaccine has been developed in several modalities including (1) an oral pill formulation (VXA-G1.1-NN) using recombinant technology to generate mucosal and systemic immune responses; (2) a plant-produced virus-like particle (VLP) vaccine (rNV-2v) without adjuvant, containing VLPs for GI.4 and GII.4; and (3) research-stage mRNA-based formulations (e.g., modified-nucleoside mRNA-LNP vaccines encoding VP1 proteins of GI.1 and GII.4). The oral formulation by Vaxart induces strong antibody responses both locally in the intestine and systemically, and the plant-produced VLP vaccine has shown robust immunogenicity and safety in early-phase clinical trials. These vaccines act by stimulating the immune system to produce neutralizing antibodies, especially targeted at blocking viral binding and infection of gastrointestinal epithelial cells. The primary indication is prevention of acute viral gastroenteritis caused by norovirus[1][2][3][5].
