A combination vaccination regimen evaluated by Aga Khan University for the prevention of poliomyelitis. This regimen incorporates bivalent oral polio vaccine (bOPV), which contains live-attenuated Sabin strains of poliovirus types 1 and 3, alongside trivalent inactivated polio vaccine (IPV) and a monovalent inactivated polio vaccine type 2 (IPV2 or mIPV2). The combination is designed to optimize population immunity against all three poliovirus serotypes, specifically addressing the immunity gap for type 2 poliovirus that occurs during the global transition from trivalent oral polio vaccine (tOPV) to bOPV. By combining mucosal immunity from the oral vaccine with systemic immunity from the inactivated vaccines, this strategy aims to prevent paralytic polio and interrupt virus transmission in endemic regions like Pakistan. The regimen was evaluated in Phase 4 clinical trials to assess its immunogenicity against type 2 poliovirus compared to traditional schedules.
