BJ-005 is a recombinant bifunctional humanized fusion protein composed of an anti-programmed death ligand 1 (PD-L1) IgG1 monoclonal antibody fused to the extracellular domain of human transforming growth factor beta receptor II (TGFβRII). The drug is designed to simultaneously inhibit two key immunosuppressive pathways in the tumor microenvironment. The PD-L1 antibody moiety binds and inhibits PD-L1 on tumor cells, blocking its interaction with PD-1 and thereby enhancing T cell-mediated antitumor immunity. The TGFβRII moiety acts as a "trap" for TGF-beta ligands, neutralizing their immunosuppressive effects. This dual mechanism aims to reduce both PD-L1– and TGF-beta–mediated immune evasion by tumors, potentially leading to increased antitumor immune responses and reduced tumor proliferation. Developed by BJ Bioscience, BJ-005 is being investigated primarily for advanced solid tumors and lymphoma[2][3][4][5].
