BK40196 is a novel small molecule dual inhibitor of c-KIT and c-Abl tyrosine kinases, which also functions as a dopamine D1 receptor agonist. It is being investigated for the treatment of Parkinson's disease and other synucleinopathies. The drug's mechanism involves reducing the levels of alpha-synuclein and neuro-inflammation while promoting protein clearance and protecting dopaminergic neurons. Preclinical studies in A53T mice have shown that BK40196 reduces microglial activity and brain tryptase levels, increases tyrosine hydroxylase-positive neurons in the substantia nigra, and improves motor behavioral performance.
