Overview

BKM120 (buparlisib) is an oral pan-class I phosphatidylinositol 3-kinase (PI3K) inhibitor developed to target the PI3K/AKT/mTOR signaling pathway, which is frequently dysregulated in cancer. Olaparib is a poly(ADP-ribose) polymerase (PARP) inhibitor that impairs DNA repair in cancer cells with homologous recombination deficiencies. The combination of BKM120 and olaparib has been investigated for synergistic antitumor effects in various cancers including breast, ovarian, gastric, and prostate cancers. Preclinical studies have shown that this combination can inhibit tumor cell proliferation and migration more effectively than either agent alone by exacerbating DNA damage and downregulating PI3K/AKT/mTOR signaling[1][5]. Clinical trials have explored this combination primarily for patients with advanced solid tumors such as ovarian and breast cancer[4][6].
