BL-1040

BL-1040 (also known as IK-5001) is an injectable, bioresorbable, alginate-based hydrogel designed to prevent pathological cardiac remodeling following an acute myocardial infarction (AMI). The liquid formulation is administered via the intracoronary artery during standard catheterization; upon reaching the infarcted myocardial tissue, it reacts with ionized calcium to form a physical scaffold. This scaffold provides mechanical support to the weakened ventricular wall, aiming to reduce wall stress, prevent infarct expansion, and mitigate the progression to congestive heart failure. Developed by BioLineRx and later licensed to Ikaria (which became Bellerophon Therapeutics), the asset was evaluated in the pivotal PRESERVATION I trial. However, the trial failed to meet its primary endpoint of improving cardiac function or clinical outcomes, leading to the cessation of its development for this indication.
