Overview

BL-2060 is an orally bioavailable small molecule inhibitor of phosphodiesterase 4 (PDE4), an enzyme that plays a critical role in the regulation of inflammatory responses by degrading cyclic adenosine monophosphate (cAMP). By inhibiting PDE4, BL-2060 increases intracellular cAMP levels, which subsequently suppresses the release of pro-inflammatory cytokines and promotes the activity of anti-inflammatory mediators. Developed by BioLineRx, the compound was primarily investigated for the treatment of chronic inflammatory airway diseases, including asthma and chronic obstructive pulmonary disease (COPD). Preclinical studies indicated that BL-2060 possessed a favorable therapeutic index compared to earlier PDE4 inhibitors, potentially offering a reduction in the gastrointestinal side effects typically associated with this drug class. However, clinical development has not progressed to late-stage trials, and the asset is no longer listed in the developer's active pipeline.
