BL-7040 is an orally available, synthetic oligonucleotide drug with dual activity on both the nervous and immune systems. It was developed primarily for the treatment of inflammatory bowel disease (IBD), including ulcerative colitis. Mechanistically, BL-7040 acts as a specific agonist of Toll-like receptor 9 (TLR9), which plays a key role in immune system modulation and inflammatory responses. Additionally, it functions as a suppressor of acetylcholinesterase, an enzyme involved in neurological pathways. This dual mechanism provides both anti-inflammatory and neuro-modulatory effects. The drug has demonstrated safety and efficacy in phase II clinical trials for ulcerative colitis but development appears to have been discontinued for this indication[1][2][3][5]. It was originally invented at the Hebrew University of Jerusalem.
