Overview

BL-8040 + pembrolizumab is a combination regimen used principally in the treatment of metastatic pancreatic ductal adenocarcinoma. BL-8040 (motixafortide) is a small molecule antagonist of the chemokine receptor CXCR4, developed to mobilize and enhance tumor infiltration by effector T cells. Pembrolizumab (Keytruda) is a monoclonal antibody targeting PD-1, functioning as a checkpoint inhibitor to enhance anti-tumor immune responses. The combination aims to synergistically modify the tumor microenvironment, promoting immune cell infiltration as well as checkpoint inhibition, in order to increase the efficacy of immunotherapy against tumors traditionally resistant to single-agent immunotherapy. This regimen is currently under clinical investigation in phase II for metastatic pancreatic cancer, often with concurrent chemotherapy[1][2][3][5].
