BL-ARC002 is a bispecific monoclonal antibody developed by Sichuan Baili Pharmaceutical (Biokin Pharmaceutical) for the treatment of locally advanced or metastatic solid tumors. It is designed to simultaneously target Programmed Cell Death 1 Ligand 1 (PD-L1) and Tumor Necrosis Factor Receptor Superfamily Member 9 (4-1BB, also known as CD137). By binding to PD-L1, BL-ARC002 acts as a checkpoint inhibitor, blocking the PD-1/PD-L1 immunosuppressive pathway. Simultaneously, it acts as a conditional agonist of 4-1BB, stimulating T-cell and NK-cell activity specifically within the tumor microenvironment where PD-L1 is expressed, thereby potentially reducing systemic toxicity associated with 4-1BB activation. The drug is currently being evaluated in Phase I clinical trials to assess its safety, tolerability, and preliminary efficacy in patients with advanced solid tumors.
