BL-B01D1 + lenvatinib is an investigational combination therapy comprising BL-B01D1, a first-in-class bispecific antibody-drug conjugate (ADC) targeting both EGFR (epidermal growth factor receptor) and HER3 (human epidermal growth factor receptor 3), and lenvatinib, an approved multi-targeted tyrosine kinase inhibitor. BL-B01D1 works by simultaneously binding to EGFR and HER3 on tumor cells, blocking proliferative and survival signaling, and delivering a cytotoxic payload upon internalization to induce cancer cell death. Lenvatinib inhibits multiple receptor tyrosine kinases involved in tumor angiogenesis and progression, including VEGFR, FGFR, PDGFRα, RET, and KIT. This combination is under clinical investigation for advanced hepatocellular carcinoma and additional solid tumor types[1][2][3][4][5][6].
