BL-M05D1 is a monoclonal antibody-drug conjugate (ADC) developed to target claudin 18.2, a protein highly expressed in certain malignancies such as gastric cancer. The drug consists of a proprietary claudin 18.2-specific antibody linked to a cytotoxic payload, enabling selective delivery of the therapeutic agent to cancer cells expressing this marker. Upon binding and internalization by the tumor cell, the ADC is trafficked to lysosomes where it releases its cytotoxic payload, inducing genotoxic stress and subsequent cancer cell death. The Fc region of the antibody is engineered for wild-type IgG function, allowing mediation of innate immune effector functions and complement-mediated cytotoxicity against targeted cells. BL-M05D1 is being evaluated as a single agent in clinical trials for patients with locally advanced or metastatic solid tumors that express claudin 18.2[1][3][5].
