Overview

BLD-1102 is a small molecule inhibitor of calpain, specifically targeting calpain-1 and calpain-2, developed by Blade Therapeutics. Calpains are calcium-dependent cysteine proteases that play a critical role in the activation of pro-fibrotic pathways, including the processing of talin and the subsequent activation of integrins and TGF-β. By inhibiting calpain activity, BLD-1102 aims to reduce the progression of fibrotic diseases such as idiopathic pulmonary fibrosis (IPF) and liver fibrosis. The compound was designed to be a potent and reversible inhibitor, offering a novel approach to treating chronic fibrotic conditions by addressing the underlying cellular mechanisms of myofibroblast activation and extracellular matrix deposition.
