Overview

ABVD is a chemotherapy combination regimen consisting of four drugs: doxorubicin (also known as Adriamycin), bleomycin, vinblastine, and dacarbazine. It is primarily used as first-line treatment for classic Hodgkin lymphoma. Each drug in the combination has a distinct mechanism of action targeting rapidly dividing cancer cells through different pathways: - Doxorubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II. - Bleomycin induces DNA strand breaks via free radical formation. - Vinblastine disrupts microtubule assembly by binding tubulin. - Dacarbazine acts as an alkylating agent causing DNA methylation and cross-linking. The regimen is administered intravenously in cycles (typically every 28 days) with two treatments per cycle. ABVD has demonstrated high efficacy in inducing remission in Hodgkin lymphoma patients and remains a standard of care due to its balance of effectiveness and manageable toxicity profile[2][3][4][5].
