Overview
Bleselumab is a fully human immunoglobulin G4 (IgG4) monoclonal antibody that acts as an antagonist of the CD40 receptor. It was developed for the prevention of organ transplant rejection and investigated for use in preventing recurrence of focal segmental glomerulosclerosis (FSGS) after kidney transplantation. Bleselumab works by blocking the interaction between CD40 and its ligand (CD40L), thereby inhibiting immune responses involved in allograft rejection and autoimmune processes. The drug was originally developed by Kyowa Kirin Pharmaceutical Development Inc., with development also involving Astellas Pharma. Clinical trials included studies in renal transplant recipients and patients at risk for recurrent FSGS, but development has been discontinued for these indications[2][3][5][7].
