Drug intelligence / Profile preview
BLN-481 (formerly BMS-986481) is a novel, preclinical-stage monoclonal antibody designed to selectively antagonize interleukin-18 receptor accessory protein (IL-18Rβ). Developed by Beeline Medicines (originally in-licensed from Bristol Myers Squibb), BLN-481 provides a receptor-specific approach to blocking IL-18-driven inflammation. Unlike ligand-directed IL-18 neutralization or IL-18 receptor alpha (IL-18Rα) blockade, which can inhibit beneficial anti-inflammatory pathways (such as IL-37 signaling), targeting IL-18Rβ selectively blocks the pathogenic, pro-inflammatory axis of IL-18 signaling while preserving beneficial biology. BLN-481 is formulated for intravenous or subcutaneous administration and is planned for clinical evaluation in healthy volunteers and patients with autoimmune and inflammatory diseases.
