BLU-222 + carboplatin is an experimental combination therapy consisting of **BLU-222**, a potent and highly selective, orally bioavailable **cyclin-dependent kinase 2 (CDK2) inhibitor**, and **carboplatin**, a DNA-damaging platinum-based chemotherapeutic. This combination is under preclinical and early clinical investigation for its ability to enhance antitumor efficacy, particularly in **CCNE1-amplified, chemotherapy-resistant ovarian and endometrial cancers**. BLU-222 acts primarily by targeting CDK2, disrupting replication fork stabilization in cancer cells with cyclin E1 dysregulation, thereby *sensitizing tumors to carboplatin-induced DNA damage*. The combination has demonstrated strong antitumor activity in preclinical models, including tumor regression in settings where either agent alone was less effective.
