**BLU-701 + carboplatin + pemetrexed** is an investigational combination regimen being evaluated for the treatment of patients with *EGFR*-mutant non-small cell lung cancer (NSCLC), particularly those with resistance mutations including C797S. BLU-701 is a next-generation, brain-penetrant, reversible EGFR tyrosine kinase inhibitor (TKI) with high potency against activating *EGFR* mutations (ex19del, L858R) and the C797S resistance mutation. Carboplatin is a platinum-based chemotherapy agent that induces DNA cross-linking leading to cell death, while pemetrexed is an antifolate chemotherapeutic that inhibits several enzymes involved in folate metabolism and DNA synthesis. The combination aims to leverage the targeted efficacy of BLU-701 with the cytotoxic effects of chemotherapy. Clinical evaluation (notably the HARMONY trial, NCT05153408) is ongoing in patients whose disease has progressed after prior EGFR-TKI therapy[1][3][7].
