BLU-808 is an investigational, highly potent and selective oral small molecule inhibitor of wild-type KIT (also known as c-KIT or stem cell growth factor receptor), developed by Blueprint Medicines. It is designed to target mast cell-driven diseases by inhibiting the activation and proliferation of mast cells through selective inhibition of wild-type KIT. Mast cells play a central role in allergic and inflammatory diseases such as chronic urticaria, allergic asthma, allergic rhinitis, allergic conjunctivitis, and mast cell activation syndrome. Preclinical studies have shown that BLU-808 leads to dose-dependent inhibition and depletion of mast cells. In Phase 1 clinical trials in healthy volunteers, the drug demonstrated robust pharmacodynamic effects with dose-dependent reductions in serum tryptase (a marker for mast cell activity) exceeding 80%, supporting its mechanism of action. The drug has a half-life suitable for once-daily oral dosing and was well-tolerated at all tested doses[1][2][4][5][6].
