BLVD101

BLVD101 (formerly DFX-1092) is a bispecific antibody designed to dually target B-cell activating factor (BAFF) and a proliferation-inducing ligand (APRIL). Developed by Boulevard Bio, a precision immunology company launched by Deerfield Management in 2026, the drug was discovered through Deerfield's 3DC drug discovery engine. By blocking both BAFF and APRIL, BLVD101 aims to inhibit key drivers of B cell-mediated autoimmune diseases. Its primary clinical focus is IgA nephropathy (IgAN), where it has demonstrated potential for quarterly (every 12 weeks) dosing based on early Phase 1 single-ascending dose data in healthy volunteers. The dual inhibition strategy is intended to reduce proteinuria and hematuria and slow disease progression in IgAN, with potential applications in other autoimmune conditions like lupus and Sjögren's disease.
