BLX-155 is a recombinant plasmin-based, direct-acting thrombolytic biologic originally developed by Biolex Therapeutics using its Lemna (LEX) expression system, intended for the rapid dissolution of blood clots in conditions such as acute peripheral arterial disease, deep vein thrombosis, and hemodialysis graft thrombosis.[1][4][16][19][21] As a recombinant form of the endogenous fibrinolytic enzyme plasmin, it is designed to directly degrade fibrin within thrombi, aiming for a potent and potentially best-in-class thrombolytic profile with improved manufacturing and pharmacologic characteristics relative to plasma-derived plasmin and indirect thrombolytics.[16] The program advanced through preclinical development at Biolex and was later transferred with the LEX platform assets to Synthon, but was subsequently discontinued in venous thrombosis indications.[5][8][21]
