BLyS-gelonin is a recombinant fusion toxin combining B lymphocyte stimulator (BLyS), a targeting ligand that selectively binds B-cell surface receptors (BR3/BAFF-R, TACI, and BCMA), with the plant-derived toxin gelonin, a type I ribosome-inactivating protein. By targeting BLyS receptors expressed almost exclusively on B cells, BLyS-gelonin allows for selective internalization into malignant B cells, where gelonin is released to inhibit protein synthesis and induce apoptosis. Preclinical results show particular cytotoxicity in B-cell malignancies such as mantle cell lymphoma, diffuse large B cell lymphoma, and B-cell precursor acute lymphoblastic leukemia. The concept has been demonstrated in both in vitro and in vivo models[1][3][7][9].
