BM011 is a first-in-class small molecule activator of the Wiskott-Aldrich Syndrome Protein (WASP), developed by BIMINI Biotech. WASP is a critical regulator of actin cytoskeleton remodeling, immune synapse formation, and vesicle trafficking. By activating WASP, BM011 induces cytoskeletal stress and forcing, which leads to mitochondrial import failure, downregulation of respiratory chain proteins, and a proteotoxic stress response. This mechanism triggers mitochondrial collapse and reveals a metabolic vulnerability in lymphoma cells, specifically involving the aldolase-dependent glycolytic pathway. Preclinical research indicates that BM011-driven WASP activation promotes apoptosis in mantle cell lymphoma models by exploiting these metabolic-cytoskeletal vulnerabilities.
