BMD-2361 is an orally bioavailable small molecule inhibitor of extracellular signal-regulated kinase 1 (ERK1) and 2 (ERK2), developed by Biomed Valley Discoveries. It targets the mitogen-activated protein kinase (MAPK) pathway, which is a critical signaling cascade involved in cell proliferation and survival. Mutations in upstream components of this pathway, such as RAS and BRAF, are common in many types of cancer and lead to constitutive activation of ERK. BMD-2361 was designed to overcome resistance mechanisms associated with BRAF and MEK inhibitors by targeting the pathway at its terminal node. Clinical development was focused on patients with advanced solid tumors harboring MAPK pathway alterations, although the primary Phase 1 study was terminated by the sponsor.
