BMP7v is an engineered, enhanced variant of Bone Morphogenetic Protein 7 (BMP7) designed to target and differentiate chemoresistant colorectal cancer stem cells (CR-CSCs). By promoting the differentiation of these stem-like cells into more mature colonic cells, BMP7v sensitizes tumors to standard chemotherapy and targeted treatments. Its mechanism involves the activation of BMP signaling, which subsequently suppresses Wnt pathway activity and reduces mesenchymal traits and survival of CR-CSCs. Additionally, BMP7v has demonstrated antiangiogenic properties in preclinical mouse models of colorectal cancer. It is particularly effective when combined with standard therapy or PI3K inhibitors, especially in tumors harboring PIK3CA mutations.
