A multi-drug combination consisting of BMS-813160, nivolumab, gemcitabine, and nab-paclitaxel. - **BMS-813160** is a potent and selective small molecule dual antagonist of CCR2 and CCR5 chemokine receptors, hypothesized to inhibit migration of inflammatory monocytes and tumor-associated macrophages, thereby reducing tumor-driven immunosuppression[3]. - **Nivolumab** is a monoclonal antibody that blocks PD-1, a checkpoint inhibitor on T cells, thereby enhancing antitumor immunity. - **Gemcitabine** (Gemzar) is an antimetabolite nucleoside analog used as a cytotoxic chemotherapeutic agent, particularly in pancreatic and other cancers. - **Nab-paclitaxel** (Abraxane) is an albumin-bound form of paclitaxel, a microtubule inhibitor, formulated for improved delivery and reduced hypersensitivity reactions. This combination is being studied in advanced and metastatic pancreatic and colorectal cancer, with the intent to leverage cytotoxic chemotherapy (gemcitabine, nab-paclitaxel) and dual immunomodulation via BMS-813160 (targeting the tumor microenvironment) and nivolumab (targeting the adaptive immune checkpoint).
