BMS-823778 + metformin is an investigational oral combination of BMS-823778, a potent and selective small molecule inhibitor of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD-1), and metformin, a well-established first-line treatment for type 2 diabetes. BMS-823778 acts by reducing intracellular activation of glucocorticoids, addressing tissue-specific glucocorticoid metabolism implicated in metabolic syndrome and type 2 diabetes. Metformin is a biguanide antihyperglycemic agent that primarily decreases hepatic glucose production and improves insulin sensitivity. The combination was considered for improving glycemic control and potentially for treating metabolic syndrome, but clinical development of BMS-823778 for related indications has been discontinued[1][2][7].
