Overview

BMS-852927 is an oral, small molecule modulator of the liver X receptor (LXR), with high selectivity for the LXRβ isoform and partial activity at LXRα. It was developed primarily for the treatment of atherosclerosis and hypercholesterolemia. The drug acts as an agonist at LXRβ, promoting cholesterol efflux and regulating lipid metabolism by inducing target genes such as ABCG1. Despite its potent activity in preclinical models, clinical development was discontinued due to increases in LDL-cholesterol and triglycerides observed in humans and primate models, along with potential safety concerns[1][2][5][7].
