Overview

BMS-866949 is a small molecule triple reuptake inhibitor (TRI), also known as a serotonin-norepinephrine-dopamine reuptake inhibitor (SNDRI), developed by Bristol Myers Squibb for the treatment of major depressive disorder (MDD). The compound is designed to simultaneously inhibit the reuptake of three key monoamine neurotransmitters—serotonin, norepinephrine, and dopamine—from the synaptic cleft. By increasing the synaptic availability of these neurotransmitters, BMS-866949 aims to provide a more comprehensive antidepressant effect and potentially address symptoms that are often resistant to selective serotonin reuptake inhibitors (SSRIs). Clinical development reached Phase 1, where its safety, pharmacokinetics, and pharmacodynamics were evaluated in healthy subjects, but the program was subsequently discontinued.
