BMS-906024 + dexamethasone is an experimental combination therapy comprising BMS-906024, a potent and selective small molecule gamma secretase inhibitor (GSI) that blocks activation of all four Notch receptors, and dexamethasone, a synthetic glucocorticoid corticosteroid. BMS-906024 is designed to inhibit the Notch signaling pathway, which is frequently dysregulated in several cancers, including hematologic malignancies such as T-cell acute lymphoblastic leukemia (T-ALL) and early T-cell precursor acute lymphoblastic leukemia (ETP-ALL). Dexamethasone exerts its cytotoxic and anti-inflammatory effects through activation of the glucocorticoid receptor, leading to cell cycle arrest and apoptosis. Preclinical and early clinical trials demonstrate that combining BMS-906024 with dexamethasone may enhance anti-leukemic effects, potentially due to GSI-induced sensitization of leukemic blasts to glucocorticoid-mediated cytotoxicity. Trials in solid tumors and combination regimens are ongoing to evaluate safety, tolerability, and efficacy[1][2][3].
