BMS-914392 is an investigational oral small molecule drug developed as a potent and selective inhibitor of the Acetylcholine-sensitive potassium channel (IKACh) current. It was designed to specifically target the atria of the heart, aiming to regulate heart contractions and maintain a constant rhythm in patients with atrial fibrillation (AF). The drug acts as a potassium channel blocker, suppressing AF without affecting ventricular refractory periods or heart rate. Clinical studies evaluated its safety, tolerability, and efficacy in reducing AF burden in patients with paroxysmal atrial fibrillation who have pacemakers. While well tolerated, clinical trials did not demonstrate significant reduction in AF episodes compared to placebo[3][4][5][6][8].
