Overview

BMS-955176 is a second-generation HIV maturation inhibitor developed by Bristol Myers Squibb for the treatment of HIV infection. It acts by inhibiting one of the final steps in the HIV lifecycle, specifically blocking protease cleavage at the CA/SP1 junction within the Gag polyprotein, which prevents proper viral maturation and results in non-infectious virus particles. Unlike first-generation maturation inhibitors such as bevirimat, BMS-955176 was designed to retain activity against viruses with naturally occurring gag polymorphisms that confer resistance to earlier drugs. The drug demonstrated potent antiviral activity in vitro and in early-phase clinical trials, including efficacy against strains resistant to other antiretrovirals. However, development of BMS-955176 has been terminated[1][3][6][7].
