BMS-955176 + dolutegravir + atazanavir + ritonavir is an investigational oral combination regimen for the treatment of HIV-1 infection. It combines BMS-955176, a second-generation maturation inhibitor that blocks the final cleavage step in the HIV-1 Gag protein and prevents production of mature, infectious virions; dolutegravir, an integrase strand transfer inhibitor (INSTI) that prevents HIV genome integration into host DNA; atazanavir, a protease inhibitor that prevents the processing of HIV polyproteins into functional viral proteins; and ritonavir, a protease inhibitor used here as a pharmacokinetic booster to increase atazanavir levels. This multi-mechanistic approach targets key stages of the HIV lifecycle, aiming to suppress viral replication, including in patients with resistance to other antiretroviral classes. Evaluated in Phase 2 trials for treatment-experienced HIV-1 patients, it is designed as a potential alternative regimen, especially for those intolerant to or failing nucleoside/nucleotide reverse transcriptase inhibitors.
