Overview

BMS-986148 is a fully human IgG1 monoclonal antibody-drug conjugate (ADC) that targets mesothelin, a tumor-associated antigen highly expressed in several solid tumors. The antibody component binds specifically to mesothelin-expressing cells, and upon internalization, delivers a cytotoxic payload—tubulysin—via a cleavable valine–citrulline linker. Tubulysin acts as a tubulin polymerization inhibitor, disrupting microtubules and inducing apoptosis in targeted cancer cells. Recent research also suggests that the tubulysin payload can induce pyroptotic cell death through cleavage of gasdermin E (GSDME), stimulating antitumor immunity. Developed by Bristol Myers Squibb, BMS-986148 has been evaluated primarily for advanced solid tumors including mesothelioma, non-small cell lung cancer, ovarian cancer, pancreatic cancer, and gastric cancer[1][2][3][4][6].
