BMS-986156 is a fully human immunoglobulin G1 (IgG1) agonist monoclonal antibody that targets the glucocorticoid-induced tumor necrosis factor receptor-related protein (GITR, also known as TNFRSF18). By binding to and activating GITR, which is expressed on various T cell subsets, BMS-986156 promotes effector T-cell activation and may reduce or inactivate regulatory T cells. This dual mechanism aims to enhance antitumor immune responses. The drug has been investigated primarily for the treatment of advanced solid tumors, both as monotherapy and in combination with immune checkpoint inhibitors such as nivolumab or ipilimumab. Developed by Bristol Myers Squibb, BMS-986156 has demonstrated pharmacodynamic activity including increased peripheral T-cell and natural killer cell proliferation but limited clinical efficacy signals in broader cancer populations[1][2][3][4][5][7].
