BMS-986172 is a highly potent, orally active, and selective inhibitor of monoacylglycerol O-acyltransferase 2 (MGAT2). It belongs to the dihydropyridinone (DHP) class of compounds and was developed as a potential treatment for metabolic disorders such as obesity and non-alcoholic steatohepatitis (NASH). Preclinical studies demonstrated that BMS-986172 significantly reduced food intake and body weight in animal models. The drug has shown favorable pharmacokinetic properties, including low effective dose projections for humans. Its mechanism-based activity was confirmed in MGAT2 knockout mice. Clinical development has reached phase 1 trials to assess safety, tolerability, pharmacokinetics, and the effect of food on absorption in healthy and obese participants[2][3][5].
