Overview

**BMS-986205 + nivolumab + chemotherapy** is a combination therapy under investigation for multiple cancers, most notably muscle-invasive bladder cancer and hepatocellular carcinoma. BMS-986205 (linrodostat), a selective oral inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1), is combined with nivolumab, a programmed cell death 1 (PD-1) immune checkpoint inhibitor monoclonal antibody, and standard chemotherapy regimens (such as gemcitabine/cisplatin, abbreviated as GC). BMS-986205 acts by inhibiting IDO1, which regulates immune response and tumor immune evasion. Nivolumab blocks PD-1 to enhance T cell-mediated anti-tumor responses. Chemotherapy targets proliferating cancer cells directly. The rationale for this combination is that targeting both immunosuppressive metabolic mechanisms (IDO1 pathway), immune checkpoint pathways (PD-1), and cell cycling (chemotherapy) may achieve additive or synergistic anti-tumor effects[1][3][4][5][9].
