BMS-986238 is an investigational, orally bioavailable macrocyclic peptide-based inhibitor of Programmed Death-Ligand 1 (PD-L1) developed by Bristol Myers Squibb. It is designed to block the PD-1/PD-L1 immune checkpoint axis, a pathway frequently exploited by cancer cells to evade immune detection. Unlike traditional monoclonal antibody checkpoint inhibitors that require intravenous administration, BMS-986238 is engineered as a macrocyclic peptide to achieve high binding affinity and metabolic stability, allowing for oral dosing. It is a successor to the first-generation peptide inhibitor BMS-986189 and is being evaluated for the treatment of solid tumors and lymphomas.
