BMS-986253 + nivolumab + ipilimumab is an experimental combination immunotherapy regimen for advanced cancers. BMS-986253 is a fully human IgG1κ monoclonal antibody that targets and binds free interleukin-8 (IL-8), inhibiting its activity and thus reducing tumor-promoting inflammation and myeloid-derived suppressor cell recruitment. Nivolumab is a programmed death-1 (PD-1) immune checkpoint inhibitor, and ipilimumab is a cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4) inhibitor; both enhance T-cell mediated antitumor immune responses. The triplet aims to improve clinical outcomes in advanced cancers, particularly in patients resistant to prior checkpoint inhibitor therapy. The combination is being studied in Phase 1 and 2 clinical trials for unresectable or metastatic solid tumors, including melanoma and other advanced cancers[1][3][4][7].
