Overview

BMS-986288 is a conditionally activated, nonfucosylated anti–CTLA-4 monoclonal antibody developed using the Probody platform. This design features a peptide mask that minimizes binding to CTLA-4 in normal tissues, aiming to reduce systemic toxicity compared to conventional anti–CTLA-4 antibodies such as ipilimumab[1][2][7]. The drug targets cytotoxic T lymphocyte antigen 4 (CTLA-4), acting as an immune checkpoint inhibitor and stimulating T lymphocyte activity against tumors[2][3][5]. It was initially developed by CytomX Therapeutics and further developed by Bristol Myers Squibb for the treatment of advanced solid tumors, including head and neck squamous cell carcinoma, non-small cell lung cancer, melanoma, triple-negative breast cancer, renal cell carcinoma, urothelial carcinoma (transitional cell carcinoma), gastric cancer, esophageal cancer, cervical cancer, and colorectal cancer[7][8]. The drug has been evaluated both as monotherapy and in combination with nivolumab in clinical trials.
